7TM Pharma develops TM30339, a NPY Y4 selective agonist to be used within gastrointestinal disorders including cancer supportive care (mucositis), malabsorphtion, Ulcerative Colitis, Crohn's disease and short bowel syndrome. 7TM Pharma has characterized the safety and tolerability of the compound in several clinical Phase I/II studies and have additionally characterized it on human colonic tissue supporting its potential clinical utilities as well as in a range of preclinical gastrointestinal disease models.
The Y4 receptor has been predicted to have significant roles in mucosal function, for example epithelial ion transport and mucus secretion, as well as modulating smooth muscle activity. mRNA for the NPY Y4 receptor has been identified in the rat proximal colon, distal colon and tissue from jejunum. Y4 receptor mRNA is present in both crypt and villus epithelia of the rat small intestine. Using selective antibodies, the Y4 receptor has been detected in goblet cells and along the basal lamina of the rat intestinal villi. Furthermore, the natural hormone for the NPY Y4 receptor, Pancreatic Polypeptide (PP), has been shown to be anti-secretory in mouse colon.
